Analysis of the toxicological profiles of cosmetic ingredients and risks for cosmetic product: feedback on the structuring and use of data within the COSMETICK digital tool

The reliability of cosmetic safety assessments depends on the quality of the toxicological profiles of ingredients. A descriptive analysis of more than 4,500 profiles, integrated into the COSMETICK digital tool, illustrates the value of centralized data management in supporting these assessments.
This study aims to identify the main toxicological concerns of cosmetic ingredients and the data gaps influencing risk assessment and the toxic potential of these same ingredients, classifying them according to their critical effects and VTRs, and comparing these data with conclusions on product safety.
The results highlight a prevalence of concerns related to skin irritation and sensitization, as well as reproductive toxicity and developmental toxicity issues. Data on genotoxicity, sensitization, and toxicity from repeated administration vary greatly, ranging from complete to partial to completely absent. These data gaps are often linked to varying regulatory requirements for REACH dossiers. The use of in silico approaches (QSAR, read-across) can help to complete these profiles.
Analysis of the available NOAELs makes it possible to characterize the distribution of the toxic potential of substances. Finally, risk assessments carried out on cosmetic products show a majority of favorable safety conclusions, with points of vigilance and risks that can be precisely identified.
This study highlights the value of a centralized, documented, and traceable digital approach to managing toxicological profiles, as well as the need for regular updates and proactive data enrichment strategies to reduce uncertainty in safety assessments.
